Poker is a minigame in Red Dead Redemption 2 (RDR2). It can be played in 5 locations as seen below. Poker becomes available after beating the Chapter 2 Main mission “Who is Not without Sin”.
Valentine
Poker can be played in the Saloon of Valentine.
Flatneck Station
Pokers becomes available at Flatneck station near your Chapter 2 camp after completing Main Mission “Who is Not without Sin” (Chapter 2). It can also be played during that particular mission.
Saint Denis
The third location is in the Saloon of Saint Denis:
Blackwater
The fourth location is in the Saloon of Blackwater (accessible when you’ve reached the Story’s Epilogue).
Tumbleweed
The fifth and last location is in the Saloon of Tumbleweed (accessible when you’ve reached the Story’s Epilogue).
You will also get to play Poker automatically in chapter 4 main mission “A Fine Night of Debauchery”. It is unmissable as part of the story. But if you want to play it any other time, head to the locations shown above.
The aim of poker is to win the pot by having the best cards at the end of the game, or by convincing other players you have the best hand so they drop out. There are lots of different card combinations that have different rankings. Refer to the game’s Help Menu > Activities > Poker, to see all the rules and card combinations.
You must play every table game for 100% completion and for the trophy “Hobby Horse”. For all other minigames check out the Red Dead Redemption 2 Minigames Guide.

It seems to me there must be a prerequisite quest to open up poker playing. I’m not far at all into chapter 2 and at the moment I can only play dominoes (which seemed to be unlocked by doing the 1st Javier quest).
On a side note a list of missions that progress the chapters would be handy somas not to void any of the missable trophies, especially the “Lending a Hand” one. Imapologise if you have already posted one but as the game and help sites are so huuuuge it’s easy to miss.

The one at Flatneck Station unlocks after “Who is Not without Sin” (Chapter 2). That’s when you play Poker in Story for the first time. Probably that’s when the others unlock, I’ll have to do some testing on old save. When I checked for the first time all locations were already available (but was after Chapter 2).
For the missions list, yes we do have one 🙂

This is correct you have to first play “Who is Not without Sin” then you can play poker, it’s the games way of introducing it to you, seems a bit daft you can’t play before then but hey ho.

You can also play poker at Blackwater, and in Tumbleweed.. and apparently you can play with your camp at your first* camp near valentine if you visit your crew at the correct time of day.

The location of the poker game in Saint Denis is on the 2nd floor of the gunsmith and not at the indicated marker upthere.
