All of the NPCs have their own questlines in Elden Ring Shadow of the Erdtree DLC. Anyone you meet will have hidden side quests for you. Just like the main game there are no waypoints or objectives. The game wants you to figure out the steps by yourself.
As a rule of thumb: you always need to exhaust all dialogues with every NPC. Some will give you hints what to do, and some will give you items related to their side quest. A few quests have multiple outcomes depending on who you side with or what items you give to an NPC.
Missable Warning: The Point of No Return is when burning the Sealing Tree at Church of the Bud (unmissable story event). This is after the Romina main boss. After this all NPCs will disappear and their questlines will fail, except Queelign, Ymir/Jolán and Igon/Priestess which are the only ones that remain doable post-story. Do all questlines before burning the Sealing Tree. Keep a backup save if possible. The questlines for Hornsent & Hornsent Grandam have to be initiated before defeating Messmer boss in the Shadow Keep. The questline for Moore has to be initiated before doing the St. Tria & Thiollier questline.
The two most important questlines are St. Trina & Thiollier and Sir Ansbach. If you do their quests you can summon them to aid you against the final story boss, which makes it much easier. Without them you’d be solo against the endboss which is extremely difficult.

The point of no return is also just progessing to a certain point in the map. Shadow Keep i think it’s called. I accidentally walked there, and then something happened to make all the NPC’s leave. No warning whatsoever. I’ve only beat 1 main boss.

Some of them move to different locations as you advance the story, but they don’t disappear completely.

Rough explanation: (Before defeating messmer collect all miquella cross and after the seal is broken(which happens once get close to shadow keep) talk to him which will advance his storyline.
After talking to both him and leda(I talked to them after giving ansbach’s letter to freyja).Leda will choose to eliminate him and hornsent on the other hand will leave highroad cross to get his revenge on messmer.
In shadow keep(forgot the location) you get option to help leda in defeating hornsent or vice versa or you can skip fight altogether by not picking any sign and just summon him during mesmer boss fight(this is the best option as you get to complete all storyline without issues) his sign should be one on the left side INSIDE the boss arena, summoning him for messmer fight will satisfy him as well as cause leda to stop pursuing him(this will result in leda setting his sight on ansbach for ansbach quest).
He attacks you later as you approach endgame his quest ends once you defeat him.

Moore is now added. Had missed his quest myself on first playthrough because I did Thiollier’s quest first.
It would actually be best to do his quest as the very first. After story I could no longer do his quest as he just repeats the same line of dialogue that he likes foraging. There are two other things that make some steps of his quest partially unavailable: advancing to Shadow Keep or doing Thiollier’s quest. It can still be done from the Point of No Return as long as Thiollier is still at Pillar Path Cross.

I have also seen that killing the forager things is bad in his quest as he has a hidden item you have to get from talking to him after healing 1 or 2 of the brood
