The Curse of the Pharaohs is the central Main Quest in Assassin’s Creed Origins’ second DLC of the same name. Until the very end, his quest doesn’t have many tasks on its own, but to complete a series of other main quests. Once these are completed, this quest also serves as this DLC’s finale.

To start this DLC and its quests, head to either Siwa (shown above and below), Alexandria or Memphis to travel to the new regions around Thebes.
After we arrive we will witness a Shadow of the Pharaoh’s attacking villagers and guards. He won’t be paying much attention to us. Fighting him actually leads to nothing, because..
..he will actually disappear back to the Afterlife, before we can finish him up.
Now we should really speak to Merti, who is the contact Amunet (Aya) gave us. Now we have seen what happens around here and want to free this region from this terrible curse.
After speaking to her, two new Main Quests will become available:

After these two, this quest will continue for a little bit.
We will meet with Isidora (who we met in a previous quest) and bring her up to speed and tell her what we have experienced.
Our little chat will be interrupted by some Roman soldiers marching in, asking questions about the priestess and her alliances.
It’s time to investigate Tychon’s office now, the guy we have taken out and obtained his ledger in an earlier quest. There are five clues to find, all on the upper floor of his house. Two of them are in a separate room, which has to be accessed from the outside, after pulling away the shelf and smashing the door, as seen below.
Seems like Bayek was not the first one to look around for clues in this house.
Now it is time for another Main Quest:
Conclusively, we are off to the final of this Quest. It is advised to be at least Level 52 to continue. Being underlevelled makes the boss-fight harder than it needs to be.
We are headed for the Temple of Karnak to end Isidora’s games of intrigues once and for all. There is a window on the southern wall where you can climb in and are almost next to her.
There will be plenty of guards around, so killing her without them noticing will be nearly impossible. You have to confirm the kill , so you have to get close to the body as well. So prepare yourself for a fight against her guards and brutes.
Once they have been dealt with and we confirm our kill, Bayek realizes that he didn’t kill her, but some random guard.
The relic in possession of Isidora must have clouded our mind.
We head a bit further on into Isidora’s quarters to find some answers. The door will be locked, but there is an opening to the right of it (as seen below).
Inside, another investigation scene will trigger and we have to find a total of six clues. Doing so will reveal what she’s been up to the whole time.
She wants to complete a ritual that her late mother told her to do, putting Amun back on his throne. Bayek tries to kill her with the hidden blade, but she morphs into the shape of Tutankhamun and our final boss-battle will begin.
Like all previous boss-fights, it is always a good idea to set fire or any other elemental damage-over-time to the enemy, because..
..we will spend a good amount of time dodging the blows and can let the elemental damage work for us in our favor in the meantime.
Once you have figured out the attack pattern and are able to land a few blows yourself, make use of your overpower-attack + to take a good portion of his health.
Eventually, you will get him down after about five minutes of dodging around and the occasional sword-strike of yours in between.
Killing him will give us our Lift the Curse-trophy and we only have to leave this afterlife and head back to the Valley of the Kings. Simply follow the glowing orb to the exit..
..which looks like the entrance to the previous afterlives we have entered so far.
This will trigger the final cutscene will Bayek and Isidora, who was finally brought to justice.
The final part of the cutscene will be with Sutekh, the first person we actually saw when we stepped into Thebes and whom we have met on a few occasions on our journey.
Bayek tasks him to get rid of the artifact which caused so much harm – especially to Sutekh himself.
This will conclude the quest-line of this DLC. Now there is still plenty to explore in Thebes as well as the Afterlives.
